產(chǎn)品活性：Gemcitabine (LY 188011) 是一種嘧啶核苷類(lèi)似物抗代謝藥 (nucleoside antimetabolite/analog) 和抗腫瘤劑。Gemcitabine 抑制 DNA 合成 (DNA synthesis) 和修復(fù)，導(dǎo)致細(xì)胞自噬 (autophagy) 和凋亡 (apoptosis)。

體外：Gemcitabine (購(gòu)自 MedChem Express，0.003-1 μM；3 天) 有效且有效地殺死小鼠和人類(lèi)衰老細(xì)胞[4]。 Gemcitabine 抑制 BxPC-3、Mia Paca-2、PANC-1、PL-45 和 AsPC-1 細(xì)胞的生長(zhǎng)，IC50 分別為 37.6、42.9、92.7、89.3 和 131.4 nM[1]。 MCE尚未獨(dú)立證實(shí)這些方法的準(zhǔn)確性。僅供參考。

體內(nèi)：Gemcitabine 可通過(guò)氣管內(nèi)噴霧給大鼠給藥，無(wú)明顯毒性，最大耐受劑量為 4 mg/kg，每周一次，持續(xù) 9 周。在 2、4 和 6 mg/kg 劑量下，Gemcitabine 通過(guò)肺部給藥的毒性低于口服給藥[2]。 在 LSL-KrasG12D/+; LSL-Trp53R172H; Pdx-1-Cre 處理過(guò)的小鼠，與安慰劑組相比，使用 Gemcitabine (50 mg/kg，ip) 或 DMAPT/Gemcitabine 組合的中位生存時(shí)間顯著增加超過(guò) 30 天 (分別為 254.5 或 255 天與 217.5 天))[3]。 MCE尚未獨(dú)立證實(shí)這些方法的準(zhǔn)確性。僅供參考。
